RESUMO

Agricultural productive services are important paths to realize the development of green agriculture, while the effect of agricultural productive services on the agricultural environment and its influencing mechanism are not yet clear. With the panel data of agricultural production in China from 2004 to 2019, by using multi-output stochastic frontier analysis with an output-oriented distance function, this study investigates agricultural environmental efficiency based on net carbon sinks. Then, this study explores the effect of agricultural productive services on agricultural environmental efficiency and its mechanisms by adopting ordinary least squares regression with fixed-effect panel model, causal steps approach, and spatial econometric method. The main findings are as follows: Firstly, agricultural productive services enhance agricultural productivity and agricultural environment by optimizing inputs and increasing outputs, and thus improve agricultural environmental efficiency. This result holds steadily after using instrumental variables to deal with endogeneity, changing the measurement of the dependent and independent variables, and subdividing the sample. Secondly, the pathways of agricultural productive services affecting agricultural environmental efficiency are mainly reflected in technology progress, planting structure adjustment, factor allocation optimization, and spatial spillover. Thirdly, due to the law of diminishing marginal returns, the impact of agricultural productive services on agricultural environmental efficiency is more significant when the level of agricultural productive services is relatively low. To improve agricultural environmental efficiency, we suggest implementing different productive service strategies in different regions, strengthening information integration, and improving infrastructure.

RESUMO

Environmental problems caused by energy consumption in the rapid popularization of China's agricultural mechanization (AM) have caused increasing concern. Using the panel data of China's 30 provinces from 2001 to 2019, this article adopts a stochastic frontier analysis method with output-oriented distance function to measure agricultural environment efficiency (AEE) based on net carbon sinks and empirically analyzes the impact of AM on AEE. The main findings are as follows: Firstly, the AEE of the nation and all provinces shows an upward trend over time and has significant spatial positive autocorrelation characteristics. Secondly, there is an inverted U-shaped relationship between AM and AEE. Meanwhile, AM has spatial spillover effect and time cumulative effect on AEE. These basic conclusions are still robust after using instrumental variables, spatial autoregressive model, sub-sample regression, changing spatial weight matrix, and independent variable. Thirdly, the effect of AM on AEE depends on the input effect and output effect caused by AM. The mechanism is mainly reflected in agricultural technology progress, expansion of the scale of farming operation, optimization of resource allocation, and spatial spillover. Given these findings, the paper adds considerable value to the empirical literature and provides various policy and practical implications.

RESUMO

Background: Radiotherapy is one of the most effective therapeutic strategies for cervical cancer patients, although radioresistance-mediated residual and recurrent tumors are the main cause of treatment failure. However, the mechanism of tumor radioresistance is still elusive. DNA damage response pathways are key determinants of radioresistance. The purpose of this study was to investigate the role and mechanism of SND1 in radioresistance of cervical cancer. Methods: A stable HeLa cell line with SND1 knockout (HeLa-KO) was generated through a modified CRISPR/Cas9 double-nicking gene editing system. The stable CaSki cell lines with SND1 knockdown (CaSki-Ctrl, CaSki-SND1-sh-1, CaSki-SND1-sh-2) were constructed through lentivirus transfection with the pSil-SND1-sh-1 and pSil-SND1-sh-2 plasmids. Results: It was observed that SND1 deficiency significantly increased the radiosensitivity of cervical cancer cells. It was also found that silencing SND1 promotes radiation-induced apoptosis. Significantly, the cells with a loss of SND1 function exhibited inefficient ataxia telangiectasia mutated pathway activation, subsequently impairing DNA repair and G2/M checkpoint arrest. In addition, threonine 103 is an important phosphorylation site of SND1 under DNA damaging stress. Conclusion: Collectively, the results of this study reveal a potent radiosensitizing effect of silencing SND1 or T103 mutation on cervical cancer cells, providing novel insights into potential therapeutic strategies for cervical cancer treatment.

RESUMO

BACKGROUND: Epidermolysis bullosa (EB) is a rare genetic disease with widely different clinical manifestations, but the relationship between genotype and phenotype is not fully understood. In the present study, we recruited a Chinese family in which two members had been diagnosed with localized EB simplex (EBS), with clinical manifestation, including blisters and erosions on the soles of the feet since infancy. OBJECTIVE: To identify and confirm the genetic variation in a Chinese family diagnosed as localized EBS. METHODS: Our study included two patients, other healthy members of the family, and 100 normal controls. Genomic DNA samples were isolated from each participant, and then polymerase chain reaction (PCR) direct sequencing was performed. RESULTS: The results of PCR direct sequencing revealed a novel heterozygous missense mutation in codon 461 of exon 7 of KRT5 (c.1382T>C), which led to an amino acid change (p.L461P) in the patients with EBS but was absent in unaffected family members and 100 unrelated control samples. CONCLUSION: The present study broadens the mutational spectrum of EBS, and this knowledge could be harnessed for prenatal screening, gene diagnosis, and gene therapy for localized EBS.

RESUMO

STUDY OBJECTIVES: The objective of the present study was to evaluate the accuracy of a home sleep apnea test (HSAT), MATRx plus (Zephyr Sleep Technologies, Calgary, Alberta, Canada), in identifying apneas and hypopneas and estimating indices of obstructive sleep apnea (OSA). METHODS: Individuals with suspected OSA underwent a one-night study wearing both HSAT and polysomnogram (PSG) sensors. The results provided by the overnight HSAT were compared with those from the simultaneously recorded PSG. The PSG data were scored manually, and the HSAT data were analyzed automatically using both preceding peak (PP) and moving average window (MW) methods for determining the reference oxyhemoglobin saturation (O2 Sat). Accuracy of the HSAT in detecting individual apneic and hypopneic events was evaluated on an epoch-by-epoch basis. The apnea-hypopnea indices from the two recording systems were compared. RESULTS: Agreement analysis for the individual apneic and hypopneic events yielded median values for sensitivity and specificity of 0.89 and 0.98 and positive and negative likelihood ratios of 37.35 and 0.11, respectively. Comparison of OSA indices between the two systems yielded correlation coefficients in the range of 0.95-0.96 and intraclass correlation coefficients ranging from 0.92-0.96. Bland-Altman analyses showed 0-2 cases lying outside the ± 2 standard deviation (SD) band and biases ranging from 2.1 to 5.3 events/h. The biases were larger for MW than PP. CONCLUSIONS: The MATRx plus HSAT identifies apneic and hypopneic events and estimates OSA indices with accuracy suitable for clinical purposes but not in children, patients with underlying lung disease, and habitual mouth-breathers. CLINICAL TRIAL REGISTRATION: Registry: ClinicalTrials.gov; Name: PSG Validation of MATRx plus AHI; Identifier: NCT03627169.

RESUMO

Through a facile-operating ultrasonic force-assisted liquid exfoliation technology, the single-layered two-dimensional (2D) [Co(CNS)2(pyz)2] n (pyz = pyrazine) nanosheets, with a thickness of sub-1.0 nm, have been prepared from the bulk precursors. The atomically thickness and the presence of abundant sulfur atoms with high electronegativity arrayed on the double surfaces of the sheets are making this kind of 2D MOF (metal-organic framework) nanosheets highly sensitive to intermolecular interactions. As a result, it can be well dispersed in all kinds of solvents to give a stable colloidal suspension that can be maintained for at least one month, accompanied by significant solvatochromic behavior and various optical properties, which thus have shown the potential to be practically applicated as in situ visual test paper for solvent identification and solvent polarity measurements. More importantly, combined with a smartphone, this kind of 2D-MOF nanosheets can be developed into in situ visual test paper to identify isomers and determine the polarity of mixed solvents quantitatively and qualitatively, suggesting the promising application of a portable, economical, and in situ visual test strategy in real world.

RESUMO

OBJECTIVE: To analyze the clinical characteristics and causative mutation in an ethnic Han Chinese family affected with punctate palmoplantar keratoderma (PPPK). METHODS: Clinical characteristics and inheritance pattern of the family were analyzed. Two seriously affected individuals from the family were investigated by whole exome sequencing. Three healthy individuals from the family and 120 non-PPPK individuals were evaluated to validate the result. RESULTS: The family was characterized by keratotic papules on the palms and soles, which gradually increased in size and number with age and coalesced with each other, particularly over the pressure part of the palms and soles. The family has featured autosomal dominant inheritance. A heterozygous frameshift variant c.419delC in exons of the CELA1 gene was identified in all affected individuals but not among non-affected members. CONCLUSION: A heterozygous frameshift variant c.419delC in CELA1 gene probably underlies the disease in the family affected with PPPK.
